Immunogenicity and Engineering of antibodies (Hervé Nozach)
The team focuses its research on the mechanisms and early stage evaluation of immunogenicity of therapeutic proteins and antibodies and on the engineering of antibodies using Yeast Surface Display. Our main objective is to develop new antibody-related molecules with high affinity, controlled selectivity and low immunogenicity.
Synthetic biology and Evolution (Oscar Pereira-Ramos)
Cells and biomolecules with new or improved function can bear considerable socio-economic impact. Our team is interested in using synthetic biology tools and concepts in order to have a deeper insight on biosystems, to create live machines and to obtain engineered molecules applied to research and health.

Job description
The position is proposed in the context of French ambition to become a leader in innovative biotherapies and bioproduction relying on a priority research equipment program (PEPR) that is co-managed by CEA and INSERM. You will participate in the engineering of immunoglobulin domains in order to improve bioproduction of bi-specific antibodies by promoting heterodimer chain pairing over homodimers. Heavy-chain/heavy-chain (CH3/CH3) and heavy-chain/ligth-chain (CH1/CL) interactions should be focused for the construction of libraries and the heterodimerization propensity of variant pairs will be evaluated using a mixed pipeline (using biotechnological and bioinformatics approaches) developed by our team and our partners. Briefly, the pipeline is designed for high throughout library-on-library screens and is based on the coupling of: i) a quantitative bacterial two-hybrid (qB2H) system that correlates protein-protein interaction (PPI) strength with cell survival; ii) NGS using long (PacBio HiFi) and short-read (Illumina) and; iii) python script for data analysis. The results should allow to get insight on promising pairs of variants that should be produced as full length antibodies in mammalian cells for validation.
Applicant Profile
MISSIONS:
- To construct variant libraries using suitable molecular biology approach (golden gate, gibson, restriction/ligation, etc)
- To conduct qB2H assays
- To prepare DNA samples for NGS
- To interpret data analysis results
- To produce promising pairs of variants in mammalian cells and to characterize them
PROFILE:
Concerning hard skills, we are looking for candidates with strong background in molecular/synthetic biology that are familiar to microbial and mammalian cell culture. Previous experience in one or more of the following domains will be advantageous: two-hybrid systems, NGS sample preparation, protein production in mammalian cells and biochemical characterization. Regarding soft skills the candidate should be organized and reliable, have critical thinking and good communication (for presentations and writing of reports or publications), be capable to team working and be motivated to bring the multidisciplinary project to the expected results. Language aptitude: English and French are convenient.
